生物活性：Etoposide (VP-16; VP-16-213) 是一種抗癌化療劑。依托泊苷抑制 拓?fù)洚悩?gòu)酶 II，從而停止 DNA 復(fù)制。依托泊苷誘導(dǎo)細(xì)胞周期停滯、細(xì)胞凋亡 和自噬^[1]。 

體外：依托泊苷能夠通過(guò) JNK/ERK 介導(dǎo)的 GSK-3 下游觸發(fā)的 RIN-m5F 細(xì)胞線粒體依賴性信號(hào)通路誘導(dǎo)細(xì)胞凋亡，從而對(duì)胰腺 β 細(xì)胞產(chǎn)生細(xì)胞毒性^[1]。依托泊苷和抗人 VEGF 顯著消除 P1 球形成能力，這是一種與該細(xì)胞亞群凋亡相關(guān)的作用^[2]。磷酸依托泊苷（0-1μM；72 小時(shí)) 以劑量依賴性方式抑制 HCT116 FBXW^+/+、FBXW^-/- 和 p53^-/-，表現(xiàn)出 IC ₅₀ 0.945 μM； 0.375 微米；和 1.437 μM，分別^[5]。依托泊苷（25 μM；6 小時(shí)）延遲 FBXW7 缺陷細(xì)胞中的 p53 恢復(fù)。此外，F(xiàn)BXW7^-/-細(xì)胞^[5]中FBXW7表達(dá)消失。

體內(nèi)：將依托泊苷 (50 μM) 和經(jīng)抗人 VEGF 處理的缺氧細(xì)胞靜脈注射到免疫缺陷小鼠體內(nèi)，顯示誘導(dǎo)肺集落的能力降低，并且潛伏期較長(zhǎng)^[2]。 Etoposide（10 mg/kg/天，靜脈注射）與 NSC 109724 和 NSC 241240 一起，減少注射了肝母細(xì)胞瘤細(xì)胞的 NMRI 裸鼠的腫瘤體積^[3]。
